医院始建于1905年美国长老会创办的“仁济医院”。民国初期至1996年前为县级医院，1997年升格为地市级医院。2003年7月10日由政府主导，本着“靠大、靠外、靠强”的原则，加盟南京鼓楼医院集团，医院更名为“南京鼓楼医院集团宿迁市人民医院”，是全国*家地市级公立医院改制医院。医院2005年9月晋升为三级综合医院，2008年12月被评为三级乙等医院。2019年2月通过三级甲等综合医院评审。2021年7月12日，更名为“南京鼓楼医院集团宿迁医院”。
